The enteric nervous system (ENS) is the largest component of the autonomic nervous system, with a complex network of neurons and glia essential for gut mobility and nutrient absorption. Hirschsprung’s disease (HSCR), the best understood enteric neuropathy, arises from ENS developmental defects during fetal stage and remains challenging to manage. Surgical resection of aganglionic segment is life-saving, but often results in chronic GI dysfunction. My project aims to develop a novel human pluripotent stem cell (hPSC)-based cell therapy for HSCR. In particular, I’m developing a hypo-immune approach that should allow grafting of human enteric precursors into animal models (eventually into patients) without requiring long-term immunosuppression.
